This variant lies in the CASR gene (transcript NM_000388.4) at coding-DNA position 2021, where A is replaced by T; at the protein level this means replaces aspartic acid at residue 674 with valine — a missense variant. Submitter rationale: This sequence change replaces aspartic acid with valine at codon 674 of the CASR protein (p.Asp674Val). The aspartic acid residue is highly conserved and there is a large physicochemical difference between aspartic acid and valine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with CASR-related conditions.
