Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006361.6(HOXB13):c.575G>A (p.Gly192Asp), citing Ambry Variant Classification Scheme 2023: The p.G192D variant (also known as c.575G>A), located in coding exon 1 of the HOXB13 gene, results from a G to A substitution at nucleotide position 575. The glycine at codon 192 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
