Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000302.4(PLOD1):c.1697G>T (p.Cys566Phe), citing Ambry Variant Classification Scheme 2023: The p.C566F variant (also known as c.1697G>T), located in coding exon 16 of the PLOD1 gene, results from a G to T substitution at nucleotide position 1697. The cysteine at codon 566 is replaced by phenylalanine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
