NM_022168.4(IFIH1):c.2575C>G (p.His859Asp) was classified as Uncertain significance for Singleton-Merten syndrome 1; Aicardi-Goutieres syndrome 7 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The aspartic acid amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals affected with IFIH1-related conditions. This sequence change replaces histidine, which is basic and polar, with aspartic acid, which is acidic and polar, at codon 859 of the IFIH1 protein (p.His859Asp). This variant is not present in population databases (gnomAD no frequency).
