NM_001110556.2(FLNA):c.1684G>A (p.Gly562Arg) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G562R variant (also known as c.1684G>A), located in coding exon 10 of the FLNA gene, results from a G to A substitution at nucleotide position 1684. The glycine at codon 562 is replaced by arginine, an amino acid with dissimilar properties. Based on data from gnomAD, the A allele has an overall frequency of <0.01% (2/181637) total alleles studied, with 1 hemizygote(s) observed. The highest observed frequency was <0.01% (1/81410) of European (non-Finnish) alleles. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
