This variant disrupts the WD40 domain of the PALB2 protein, which is essential for PALB2's function in DNA repair (PMID: 19423707, 16793542, 19609323). While functional studies have not been performed to directly test the effect of this variant on PALB2 protein function, this suggests that disruption of this region of the protein is causative of disease. This variant has not been reported in the literature in individuals with PALB2-related conditions. This variant is a gross deletion of the genomic region encompassing exon(s) 2-12 of the PALB2 gene. While this deletion is not anticipated to lead to nonsense mediated decay, it is expected to disrupt the C-terminus of the protein.
