Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_002528.7(NTHL1):c.167T>C (p.Leu56Pro), citing Ambry Variant Classification Scheme 2023: The p.L64P variant (also known as c.191T>C), located in coding exon 2 of the NTHL1 gene, results from a T to C substitution at nucleotide position 191. The leucine at codon 64 is replaced by proline, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
