Uncertain significance for Diffuse cerebral and cerebellar atrophy - intractable seizures - progressive microcephaly syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005051.3(QARS1):c.455A>C (p.Glu152Ala), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the QARS1 gene (transcript NM_005051.3) at coding-DNA position 455, where A is replaced by C; at the protein level this means replaces glutamic acid at residue 152 with alanine — a missense variant. Submitter rationale: This sequence change replaces glutamic acid with alanine at codon 152 of the QARS protein (p.Glu152Ala). The glutamic acid residue is highly conserved and there is a moderate physicochemical difference between glutamic acid and alanine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with QARS-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
