This variant lies in the NF2 gene (transcript NM_000268.4) at coding-DNA position 1320, where G is replaced by A; at the protein level this means replaces methionine at residue 440 with isoleucine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This sequence change replaces methionine with isoleucine at codon 440 of the NF2 protein (p.Met440Ile).
