Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000222.3(KIT):c.2651A>G (p.Lys884Arg), citing Ambry Variant Classification Scheme 2023: The p.K884R variant (also known as c.2651A>G), located in coding exon 19 of the KIT gene, results from an A to G substitution at nucleotide position 2651. The lysine at codon 884 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
