Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001963.6(EGF):c.1480C>T (p.Arg494Ter), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Arg494*) in the EGF gene. It is expected to result in an absent or disrupted protein product. However, the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in EGF cause disease. This variant is present in population databases (rs753726274, gnomAD 0.007%). This variant has not been reported in the literature in individuals affected with EGF-related conditions. ClinVar contains an entry for this variant (Variation ID: 1426041). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site.
