Uncertain significance for Joubert syndrome; Meckel-Gruber syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001082538.3(TCTN1):c.667C>T (p.Pro223Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces proline with serine at codon 223 of the TCTN1 protein (p.Pro223Ser). The proline residue is highly conserved and there is a moderate physicochemical difference between proline and serine. This variant is present in population databases (rs770297289, ExAC 0.001%). This variant has not been reported in the literature in individuals affected with TCTN1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
