Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_024818.6(UBA5):c.446A>G (p.Tyr149Cys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces tyrosine with cysteine at codon 149 of the UBA5 protein (p.Tyr149Cys). The tyrosine residue is moderately conserved and there is a large physicochemical difference between tyrosine and cysteine. This variant has not been reported in the literature in individuals affected with UBA5-related conditions. This variant is present in population databases (rs376247071, ExAC 0.01%). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Not Available"; PolyPhen-2: "Benign"; Align-GVGD: "Not Available").
