Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000234.3(LIG1):c.1715A>G (p.Gln572Arg), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals affected with LIG1-related conditions. This variant is present in population databases (rs764577674, gnomAD 0.006%). This sequence change replaces glutamine, which is neutral and polar, with arginine, which is basic and polar, at codon 572 of the LIG1 protein (p.Gln572Arg). ClinVar contains an entry for this variant (Variation ID: 1424768). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive.
