Uncertain significance for Hereditary nonpolyposis colorectal neoplasms — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000249.4(MLH1):c.208A>G (p.Lys70Glu), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the MLH1 gene (transcript NM_000249.4) at coding-DNA position 208, where A is replaced by G; at the protein level this means replaces lysine at residue 70 with glutamic acid — a missense variant. Submitter rationale: This sequence change replaces lysine, which is basic and polar, with glutamic acid, which is acidic and polar, at codon 70 of the MLH1 protein (p.Lys70Glu). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with MLH1-related conditions. ClinVar contains an entry for this variant (Variation ID: 1424678). Invitae Evidence Modeling incorporating data from in vitro experimental studies (internal data) did not meet the statistical confidence thresholds required to predict the impact of this variant on MLH1 function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
