Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000062.3(SERPING1):c.988T>G (p.Tyr330Asp), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces tyrosine with aspartic acid at codon 330 of the SERPING1 protein (p.Tyr330Asp). The tyrosine residue is highly conserved and there is a large physicochemical difference between tyrosine and aspartic acid. This variant is present in population databases (rs532337986, ExAC 0.009%). This variant has been observed in individual(s) with hereditary angioedema (PMID: 29753808; Invitae). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0").
