Uncertain significance for Mowat-Wilson syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014795.4(ZEB2):c.3608C>T (p.Ser1203Leu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine, which is neutral and polar, with leucine, which is neutral and non-polar, at codon 1203 of the ZEB2 protein (p.Ser1203Leu). This variant is present in population databases (rs758670673, gnomAD 0.0009%). This variant has not been reported in the literature in individuals affected with ZEB2-related conditions. ClinVar contains an entry for this variant (Variation ID: 1424498). Invitae Evidence Modeling incorporating data from in vitro experimental studies (internal data) indicates that this missense variant is not expected to disrupt ZEB2 function with a negative predictive value of 95%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
