Submitter rationale: This sequence change replaces glycine, which is neutral and non-polar, with glutamic acid, which is acidic and polar, at codon 1189 of the COL4A3 protein (p.Gly1189Glu). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with COL4A3-related conditions (PMID: 35386907, 35422838; internal data). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 1424247). Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown.
