Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_003036.4(SKI):c.340G>A (p.Val114Met), citing Ambry Variant Classification Scheme 2023: The c.340G>A (p.V114M) alteration is located in exon 1 (coding exon 1) of the SKI gene. This alteration results from a G to A substitution at nucleotide position 340, causing the valine (V) at amino acid position 114 to be replaced by a methionine (M). Based on data from gnomAD, the A allele has an overall frequency of 0.003% (1/31330) total alleles studied. The highest observed frequency was 0.012% (1/8692) of African alleles. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
