This sequence change replaces serine, which is neutral and polar, with alanine, which is neutral and non-polar, at codon 1127 of the CPLANE1 protein (p.Ser1127Ala). This variant is present in population databases (rs776423792, gnomAD 0.003%). ClinVar contains an entry for this variant (Variation ID: 1423771). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt CPLANE1 protein function. This variant disrupts the p.Ser1127 amino acid residue in CPLANE1. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 24178751, 31158925).
