Uncertain significance for Aortic valve disease 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005585.5(SMAD6):c.277A>C (p.Met93Leu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces methionine with leucine at codon 93 of the SMAD6 protein (p.Met93Leu). The methionine residue is weakly conserved and there is a small physicochemical difference between methionine and leucine. The frequency data for this variant in the population databases is considered unreliable, as metrics indicate insufficient coverage at this position in the ExAC database. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
