Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001374736.1(DST):c.21269G>A (p.Arg7090Gln), citing Ambry Variant Classification Scheme 2023: The p.R4971Q variant (also known as c.14912G>A), located in coding exon 83 of the DST gene, results from a G to A substitution at nucleotide position 14912. The arginine at codon 4971 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
