NM_030777.4(SLC2A10):c.643C>T (p.Arg215Trp) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R215W variant (also known as c.643C>T), located in coding exon 2 of the SLC2A10 gene, results from a C to T substitution at nucleotide position 643. The arginine at codon 215 is replaced by tryptophan, an amino acid with dissimilar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
