Uncertain significance for Cranioectodermal dysplasia 2; Short-rib thoracic dysplasia 7 with or without polydactyly — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_020779.4(WDR35):c.3340G>A (p.Glu1114Lys), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the WDR35 gene (transcript NM_020779.4) at coding-DNA position 3340, where G is replaced by A; at the protein level this means replaces glutamic acid at residue 1114 with lysine — a missense variant. Submitter rationale: This sequence change replaces glutamic acid with lysine at codon 1125 of the WDR35 protein (p.Glu1125Lys). The glutamic acid residue is weakly conserved and there is a small physicochemical difference between glutamic acid and lysine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with WDR35-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
