Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000245.4(MET):c.2242C>A (p.His748Asn), citing Ambry Variant Classification Scheme 2023: The p.H748N variant (also known as c.2242C>A), located in coding exon 8 of the MET gene, results from a C to A substitution at nucleotide position 2242. The histidine at codon 748 is replaced by asparagine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
