The aspartic acid amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. This variant has not been reported in the literature in individuals affected with IL12RB1-related conditions. This variant is present in population databases (rs573390658, ExAC 0.02%). This sequence change replaces glycine with aspartic acid at codon 167 of the IL12RB1 protein (p.Gly167Asp). The glycine residue is moderately conserved and there is a moderate physicochemical difference between glycine and aspartic acid.
