This variant lies in the RTEL1 gene (transcript NM_001283009.2) at coding-DNA position 2137, where C is replaced by G; at the protein level this means replaces histidine at residue 713 with aspartic acid — a missense variant. Submitter rationale: This sequence change replaces histidine with aspartic acid at codon 713 of the RTEL1 protein (p.His713Asp). The histidine residue is moderately conserved and there is a moderate physicochemical difference between histidine and aspartic acid. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with RTEL1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated.
