This variant lies in the TSC2 gene (transcript NM_000548.5) at the canonical splice donor site of the intron immediately after coding-DNA position 2966, where G is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: The c.2966+1G>A intronic variant results from a G to A substitution one nucleotide after coding exon 25 of the TSC2 gene. This nucleotide position is highly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice donor site. Alterations that disrupt the canonical splice site are expected to cause aberrant splicing.
