Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001330360.2(POLA1):c.2138A>G (p.His713Arg), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). This sequence change replaces histidine with arginine at codon 707 of the POLA1 protein (p.His707Arg). The histidine residue is highly conserved and there is a small physicochemical difference between histidine and arginine. This variant is present in population databases (rs752900734, ExAC 0.002%). This variant has not been reported in the literature in individuals with POLA1-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
