This variant lies in the PMS2 gene (transcript NM_000535.7) at coding-DNA position 75, where G is replaced by C; at the protein level this means replaces glutamine at residue 25 with histidine — a missense variant. Submitter rationale: In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This sequence change replaces glutamine with histidine at codon 25 of the PMS2 protein (p.Gln25His). The glutamine residue is highly conserved and there is a small physicochemical difference between glutamine and histidine. This variant is not present in population databases (ExAC no frequency).
