Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001048174.2(MUTYH):c.1030T>C (p.Ser344Pro), citing Ambry Variant Classification Scheme 2023: The p.S372P variant (also known as c.1114T>C), located in coding exon 12 of the MUTYH gene, results from a T to C substitution at nucleotide position 1114. The serine at codon 372 is replaced by proline, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
