This variant lies in the PTEN gene (transcript NM_000314.8) at coding-DNA position 11, where T is replaced by C; at the protein level this means replaces isoleucine at residue 4 with threonine — a missense variant. Submitter rationale: This missense variant replaces isoleucine with threonine at codon 4 of the PTEN protein. Computational prediction tool suggests that this variant may not impact protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with PTEN-related disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
