NM_007194.4(CHEK2):c.1174G>C (p.Ala392Pro) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Autosomal Dominant and X-Linked criteria (10/2015): Ã¢â‚¬â€¹The p.A392P variant (also known as c.1174G>C) is located in coding exon 10 of the CHEK2 gene. This alteration results from a G to C substitution at nucleotide position 1174. The alanine at codon 392 is replaced by proline, an amino acid with similar properties. This variant was not reported in population-based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP) and 1000 Genomes Project. To date, this alteration has been detected with an allele frequency of approximately 0.01% (greater than 8000 alleles tested) in our clinical cohort (includes this individual). Based on protein sequence alignment, this amino acid position is completely conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of p.A392P remains unclear.
