This variant lies in the MYH2 gene (transcript NM_017534.6) at coding-DNA position 5260, where G is replaced by T; at the protein level this means replaces alanine at residue 1754 with serine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. This variant has not been reported in the literature in individuals affected with MYH2-related conditions.
