NM_006361.6(HOXB13):c.358T>C (p.Tyr120His) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Y120H variant (also known as c.358T>C), located in coding exon 1 of the HOXB13 gene, results from a T to C substitution at nucleotide position 358. The tyrosine at codon 120 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
