Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001080467.3(MYO5B):c.4459G>A (p.Asp1487Asn), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals affected with MYO5B-related conditions. This sequence change replaces aspartic acid, which is acidic and polar, with asparagine, which is neutral and polar, at codon 1487 of the MYO5B protein (p.Asp1487Asn). This variant also falls at the last nucleotide of exon 33, which is part of the consensus splice site for this exon. This variant is present in population databases (rs200737204, gnomAD 0.05%). ClinVar contains an entry for this variant (Variation ID: 1419432). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant¬† is likely to be tolerated. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
