Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 2e-05 in 251484 control chromosomes, predominantly at a frequency of 0.00012 within the Latino subpopulation in the gnomAD database. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.1423G>A has been reported in the literature in individuals affected with breast cancer and in unaffected controls (example, Momozawa_2018, Weitzel_2019) and as a presumed somatic variant in small intestinal-type high-grade adenoma (example, Ota_2020). The variant was also reported only in the control cohort, not in the case cohort in a large case-control study for Biliary tract cancer (Okawa_2023). These report(s) do not provide unequivocal conclusions about association of the variant with Breast Cancer. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
