Uncertain significance for Facioscapulohumeral muscular dystrophy 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_015295.3(SMCHD1):c.1150G>T (p.Gly384Trp), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals affected with SMCHD1-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces glycine with tryptophan at codon 384 of the SMCHD1 protein (p.Gly384Trp). The glycine residue is highly conserved and there is a large physicochemical difference between glycine and tryptophan. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0").
