Uncertain significance for Combined immunodeficiency due to ORAI1 deficiency; Myopathy, tubular aggregate, 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000012.12:g.121641254G>A, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamic acid, which is acidic and polar, with lysine, which is basic and polar, at codon 173 of the ORAI1 protein (p.Glu173Lys). This variant is present in population databases (rs533220170, gnomAD 0.04%). This variant has not been reported in the literature in individuals affected with ORAI1-related conditions. ClinVar contains an entry for this variant (Variation ID: 1418108). Algorithms developed to predict the effect of variants on gene product structure and function are not available or were not evaluated for this variant. Experimental studies have shown that this missense change does not substantially affect ORAI1 function (PMID: 33361160).
