Uncertain significance for Hereditary breast ovarian cancer syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_007294.4(BRCA1):c.5470A>G (p.Ile1824Val), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the BRCA1 gene (transcript NM_007294.4) at coding-DNA position 5470, where A is replaced by G; at the protein level this means replaces isoleucine at residue 1824 with valine — a missense variant. Submitter rationale: This sequence change replaces isoleucine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 1824 of the BRCA1 protein (p.Ile1824Val). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with BRCA1-related conditions. ClinVar contains an entry for this variant (Variation ID: 141804). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. Experimental studies have shown that this missense change does not substantially affect BRCA1 function (PMID: 30209399). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
