This variant lies in the CDH1 gene (transcript NM_004360.5) at coding-DNA position 325, where A is replaced by C; at the protein level this means replaces lysine at residue 109 with glutamine — a missense variant. Submitter rationale: The CDH1 c.325A>C (p.K109Q) variant has not been reported in the literature to our knowledge. It was observed in 1/15416 chromosomes of the Non-Finnish European (NFE) subpopulation in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). This variant has been reported in ClinVar (Variation ID 141801). Functional studies have not been performed, and in silico predictions of the variant's effect on protein function are inconclusive. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
