Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001171.6(ABCC6):c.1742A>G (p.Gln581Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamine with arginine at codon 581 of the ABCC6 protein (p.Gln581Arg). The glutamine residue is weakly conserved and there is a small physicochemical difference between glutamine and arginine. This variant is not present in population databases (ExAC no frequency). This missense change has been observed in individual(s) with clinical features of pseudoxanthoma elasticum (Invitae). In at least one individual the data is consistent with the variant being in trans (on the opposite chromosome) from a pathogenic variant. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
