NM_000465.4(BARD1):c.1298A>G (p.His433Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H433R variant (also known as c.1298A>G), located in coding exon 4 of the BARD1 gene, results from an A to G substitution at nucleotide position 1298. The histidine at codon 433 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
