Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001235.5(SERPINH1):c.788A>G (p.Lys263Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces lysine with arginine at codon 263 of the SERPINH1 protein (p.Lys263Arg). The lysine residue is highly conserved and there is a small physicochemical difference between lysine and arginine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with SERPINH1-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt SERPINH1 protein function.
