Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001034116.2(EIF2B4):c.1249C>T (p.Arg417Trp), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine, which is basic and polar, with tryptophan, which is neutral and slightly polar, at codon 416 of the EIF2B4 protein (p.Arg416Trp). This variant is present in population databases (rs759071387, gnomAD 0.003%). This missense change has been observed in individual(s) with clinical features of leukoencephalopathy with vanishing white matter (PMID: 20016818). ClinVar contains an entry for this variant (Variation ID: 1417167). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
