Uncertain significance for Combined immunodeficiency due to ZAP70 deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001079.4(ZAP70):c.202A>T (p.Thr68Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces threonine with serine at codon 68 of the ZAP70 protein (p.Thr68Ser). The threonine residue is moderately conserved and there is a small physicochemical difference between threonine and serine. This variant is present in population databases (rs138628302, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with ZAP70-related conditions. ClinVar contains an entry for this variant (Variation ID: 1416285). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
