For these reasons, this variant has been classified as Pathogenic. ClinVar contains an entry for this variant (Variation ID: 1416115). This missense change has been observed in individual(s) with multiple acyl-CoA dehydrogenase deficiency (PMID: 30022752). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces asparagine, which is neutral and polar, with isoleucine, which is neutral and non-polar, at codon 331 of the ETFDH protein (p.Asn331Ile).
