Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000477.7(ALB):c.272A>G (p.His91Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces histidine with arginine at codon 91 of the ALB protein (p.His91Arg). The histidine residue is moderately conserved and there is a small physicochemical difference between histidine and arginine. This variant is present in population databases (rs773096498, gnomAD 0.003%). This variant has not been reported in the literature in individuals affected with ALB-related conditions. ClinVar contains an entry for this variant (Variation ID: 1416088). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
