This variant lies in the TP53 gene (transcript NM_000546.6) at the canonical splice donor site of the intron immediately after 29 bases upstream of the translation start (5' untranslated region), where G is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This sequence change falls in intron 1 of the TP53 gene. It does not directly change the encoded amino acid sequence of the TP53 protein. It affects a nucleotide within the consensus splice site. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with TP53-related conditions. ClinVar contains an entry for this variant (Variation ID: 1415968).
