Submitter rationale: Variant summary: PRODH c.1A>G (p.Met1Val) alters the initiation codon and is predicted to result either in absence of the protein or truncation of the encoded protein due to translation initiation at a downstream codon. The nearest downstream in-frame Met occurs in codon 109, which is the start codon in an alternate transcript. No pathogenic missense or in-frame indel variants have been found upstream of this alternate start codon. Two of three in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 3.2e-05 in 31148 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance.
